Prescott Island is the largest of five islands that are located along the eastern side of Prince of Wales Island. The five islands — Pandora (3.7 km to the south), Prescott, Vivian (3.7 km to the north), Lock, and Binstead — form a barrier across the entrance in to Browne Bay. The waters to the east of Prescott Island are those of the Peel Sound — separating Prince of Wales Island from Somerset Island (at right edge of the image). The outer islets of Somerset Island's western coast are situated just 23 km to the east.

The island has a surface area of 412 km², is oval in shape, measuring 30 km in length and 18 km in width.
